Oxytocin is a cyclic nonapeptide hormone with an intramolecular disulfide bridge between the cysteine residues at positions 1 and 6, forming the six-residue ring that defines the neurohypophyseal peptide family.
Oxytocin is a cyclic nonapeptide hormone with an intramolecular disulfide bridge between the cysteine residues at positions 1 and 6, forming the six-residue ring that defines the neurohypophyseal peptide family.
It was the first polypeptide hormone to be chemically synthesised, work that earned Vincent du Vigneaud the 1955 Nobel Prize in Chemistry, and it remains a standard reference compound in peptide chemistry.
The disulfide bridge is the critical handling consideration. Reducing agents will open the ring and destroy activity, so any buffer containing DTT or beta-mercaptoethanol is incompatible. Its reconstituted shelf life is shorter than most of the catalogue.

Vials arrive lyophilized and sealed under nitrogen. Bring the vial fully to room temperature before breaking the seal — opening a cold vial condenses atmospheric moisture onto the cake.
Direct diluent gently against the inside wall of the vial rather than onto the cake, and allow the material to dissolve without agitation. Vortexing shears longer sequences and introduces air.
Lyophilized: -20 C, desiccated. Reconstituted: 2-8 C, use within 14 days.
A full walkthrough is in the reconstitution guide, and the calculator will work out concentration and draw volume for this vial size.
Standard laboratory practice applies: appropriate personal protective equipment, no consumption, no administration to humans or animals, and disposal in accordance with institutional protocol and local regulation.

The disulfide bridge is susceptible to reduction and to disulfide scrambling in solution over time.
Anything containing a reducing agent - DTT, beta-mercaptoethanol, TCEP - will open the ring.
They share the cyclic scaffold but differ at positions 3 and 8, which is what separates their receptor selectivity.
