Cagrilintide is a 32-residue synthetic analogue of amylin, the pancreatic hormone co-secreted with insulin, carrying a C16 fatty diacid side chain for albumin binding.
Cagrilintide is a 32-residue synthetic analogue of amylin, the pancreatic hormone co-secreted with insulin, carrying a C16 fatty diacid side chain for albumin binding.
Native human amylin is notoriously prone to forming amyloid fibrils, and the substitutions in this analogue are specifically designed to suppress that aggregation - which is why the synthetic analogue is workable at bench concentrations where native amylin is not.
The lipidation makes it comparatively hydrophobic. A mildly alkaline buffer will dissolve it considerably faster than neutral water, and material should not be left as a concentrated solution at room temperature.

Vials arrive lyophilized and sealed under nitrogen. Bring the vial fully to room temperature before breaking the seal — opening a cold vial condenses atmospheric moisture onto the cake.
Direct diluent gently against the inside wall of the vial rather than onto the cake, and allow the material to dissolve without agitation. Vortexing shears longer sequences and introduces air.
Lyophilized: -20 C, desiccated. Reconstituted: 2-8 C, use within 30 days.
A full walkthrough is in the reconstitution guide, and the calculator will work out concentration and draw volume for this vial size.
Standard laboratory practice applies: appropriate personal protective equipment, no consumption, no administration to humans or animals, and disposal in accordance with institutional protocol and local regulation.

Native human amylin aggregates into amyloid fibrils readily. The substitutions in this analogue suppress that.
It binds serum albumin, which is the basis of the analogue's extended half-life.
A mildly alkaline buffer. The lipid side chain makes it relatively hydrophobic in neutral water.
